Tumor necrosis factor α converting enzyme (TACE) is a metalloprotease initially identified by its ability to cleave the membrane-anchored, 26 kDa form of TNF-α and release the soluble, 17 kDa form which induces inflammation. It has subsequently been shown to be a major sheddase of many other membrane proteins including TGF-α, p75 and p55 receptors, L-selectin, Growth Hormone Receptor, Amyloid Precursor Protein, Notch, IL-6 and Type II IL-1 receptors, fractalkine, ErbB4, neuregulin, and LDL receptor. The mature form of TACE is found on the cell surface and intracellularly in virtually every tissue and its protease activity is inhibited by TIMP-1, -2, -3, and -4. This assay has a sensitivity of 0.16 ng/ml, a dynamic range from 0.625 to 20 ng/ml, and is fully validated for human cell lysates.
