Kinase Activity Assays

QTL Lightspeed Kinase Activity Assays are a universal mix and measure product that does not require antibodies or radioactive labels. The technology combines sensitivity with speed for high-throughput screening of kinases, based on fluorescence "superquenching" technology. The homogeneous one-step "mix and measure" process requires no washing steps, no radioactive compounds, no antibodies and no custom instrumentation. The kits work with either peptide or protein substrates and are validated for miniaturization in 96- and 384-well plate formats. The detection platform is based on fluorescence-superquenching of a QTL Sensor, which is comprised of highly fluorescent conjugated polymers co-located with phosphate coordinating metal ions on the surface of microspheres. As kinase substrates, peptides are used that contain a fluorescence quencher within their sequence. After the kinase phosphorylates the peptide substrate the phosphorylated product binds to the QTL Sensor resulting in superquenching of polymer fluorescence. The decrease in fluorescence is a measure of kinase enzymatic activity. Since the QTL assay detects phosphorylated serine, threonine and tyrosine residues, the researcher is presented with a powerful and generic tool to evaluate phosphorylation. When used with the calibrator peptide (included and specific for each kinase assay) in the endpoint measurement kit, the researcher is able to generate standard curves and determine the amount of sample phosphorylation.
