HPLC Column Temperature Controller
Polaratherm Total Temperature Controller can heat HPLC column packings to elevated temperatures without causing column bleed or degradation. Column bleed is a problem as it leaches detectable compounds from the column packing materials causing interference with detection of the compounds of interest. To date silica based column packings have only been stable to temperatures of 60 C. In a new application note Selerity has shown that there are a number of columns available made from varying materials, such as zirconia, that can withstand higher temperatures without the effects of degradation. The columns used in the separations were from a number of manufacturers. Column leaching is also evidence of degradation of the stationary phase. Polaratherm was able to heat the Selerity Blaze C8 column(which is silica based), the Hamilton PRP-1 and the Thermo Electron Hypercarb to temperatures of up to 200 C, depending on the columns base, without any evidence of column bleed. The results were obtained by evaluating a number of different stationary phases under temperature programmed conditions to assess the amount of column bleed observed with increasing temperature. An innovative preheater design eliminates band broadening. The preheater assembly is calibrated to the oven temperature upon installation to ensure that the mobile phase temperature closely follows that of the column compartment. The Polaratherm is compatible and can be seamlessly integrated with any existing high performance liquid chromatograph.
