Antibody Conjugation Kit
This kit allows conjugation of antibodies to Qdot 655 nanocrystals. The kit contains all necessary reagents and components for two conjugation reactions, each using 300 μg of IgG class antibody, as well as a detailed protocol.
The conjugation uses a well-known chemistry based on the fast and efficient coupling of thiols to maleimide groups. The starting materials provided with the kit are chemically derivatized Qdot 655 nanocrystals containing a number of amine groups on their surface. The first step in the conjugation process is the conversion of these amines to thiol-reactive maleimide groups. This is achieved easily with the hetero-bifunctional crosslinker 4-(maleimidomethyl)-1-cyclohexanecarboxylic acid N-hydroxysuccinimide ester (SMCC). Following a 60 min reaction, the excess crosslinker is removed from the activated quantum dots by gel filtration chromatography.
