Avian IgY Antibody Conjugates For Proteomics Sample Processing
Charles River Proteomic Services Inc. (Worcester, MA) and GenWay Biotech Inc. (San Diego, CA) have collaborated to produce avian IgY polyclonal antibodies that function as a fractionation step prior to protein separation. The antibodies are covalently conjugated to microbead carriers to provide immunoaffinity separation from depletions of serum and plasma samples. While the availability and proteomic content of plasma and serum samples makes these matrices of particular interest, the large dynamic range of proteins (10xxx10 - 10xxx12) presents challenges to the accessibility of lower abundance proteins. By applying this technology in a fractionation step prior to protein separation, This technology is a sample processing step prior to proteomic analysis. It specifically removes abundant proteins from serum and plasma that would otherwise interfere with detection of rare, but important protein biomarkers. As a result of the collaboration, the two companies have developed low- cost aviaian antibodies with high antigen specificity and binding capacity. Conjugated anti-Human Serum Albumin (HSA), anti-IgG, anti-Transferrin, anti-Fibrinogen, anti-IgA, and anti-IgM are the first of many important protein immunoaffinity separation products.
