New iProof high-fidelity DNA polymerase incorporates a nonspecific DNA-binding domain (Sso7d) that imparts a sliding grip on the minor groove of the template DNA. This lends physical stability to the polymerase-DNA complex, enhancing processing abilities and enabling fast, accurate replication of templates. The enhanced processing ability reduces extension steps (to 15–30 sec/kb) and overall run times (to 25–35 min), and it enables the use of less enzyme (0.25–1 U/reaction). Fusing the double-stranded DNA binding protein Sso7d to iProof polymerase gives it a powerful sliding grip on replicated DNA.
