Expression of Firefly Luciferase After Delivery Of A Luciferase mRNA
The indicated cell lines were plated in 12-well plates and incubated overnight at 37 C. They were then transfected at approximately 80% confluency with 1.0 μg of a capped, polyadenylated in vitro transcript encoding firefly luciferase using the TransIT-mRNA Transfection Kit. Approximately 18 hours post-transfection, the cells were harvested, lysed and the amount of luciferase activity in each lysate was determined using a standard luciferase assay and a Berthold Centro LB 960 plate luminometer. Bars indicate the total average amount of luciferase activity present in each well of the 12-well plate in two or three independent experiments.
