cDNA Clones
The yeast shuttle vector pORF-CLONE is an intelligent method for an easy and cost-effective selection of clones containing long cDNA inserts with a correct open reading frame (ORF). The background of false ORFs can be significantly reduced, allowing for the generation of expression libraries enriched for clones with correct ORFs producing authentic polypeptides. An optimal expression of soluble, post-translational modified recombinant proteins is guaranteed by the vector-own, inducible CUP1 promoter and the eukaryotic Saccharomyces cerevisiae host system. Genes cloned in pORF-CLONE are expressed in S. cerevisiae as fusion proteins with HIS- and HA-tags enabling a simple, fast and reproducible purification via affinity chromatography and detection by commercially available specific antibodies.
