Reduced Serum Media Formulations

Gibco Advanced D-MEM/F-12 and Advanced RPMI 1640 reduced serum media formulations enable researchers involved in mammalian cell culture to reduce Fetal Bovine Serum (FBS) supplementation by 50 to 90 percent with little or no change in cell growth, promotion, morphology or function. The media are standard basal formulations of the classical Dulbecco's Modified Eagle Medium: Nutrient Mixture F-12 (D-MEM/F-12) and RPMI 1640, enriched with additional ingredients, all of which are normal constituents of serum. Cells normally grown in classical media with 5-20 percent serum will grow in Advanced D-MEM/F-12 or Advanced RPMI 1640 with only 0.5-2 percent FBS. The effort that is typically spent in qualifying new lots is saved due to the decrease in the percentage of serum concentration, which increases the lifespan of serum stock proportionately. Additionally, there are reduced amounts of undefined proteins. Where there are fewer lot-to-lot changes in FBS, there are fewer chances of experimental variability. Available in a 1× liquid format, the media only require supplementation with L-glutamine or GlutaMAX-I, and the appropriate reduced amount of serum. For most applications, no weaning of adaptation procedures are necessary to reduce serum supplementation by at least 50 percent in a wide variety of cell lines. Greater reduction can be achieved with minimal adaptation. The serum concentration reductions achieved with these Advanced formulation will vary with the cell line and basal medium used.
