Gene Selection To Validated Assay

Pentadot Plus™, from High Throughput Genomics, enables rapid and accurate quantification of RNA expression levels in cells treated with potential new drugs. It is based on HTG's ArrayPlate technology, which facilitates simultaneous measurement of RNA expression levels in hundreds of active genes and, by extension, provides critical information about gene expression and protein function. Pentadot is comprised of five dots, oligonucleotides, printed at the bottom of each well of a user-programmable, 96-well plate. Each well includes one user-defined target, three housekeeping genes and one procedural control. Wells can measure the same target gene or a different target. Plates are programmed using standard reagents created on-site, with no DNA extraction, reverse transcription or PCR required. Researchers can go from gene selection to a validated assay in a week. The kit contains reagent design and imaging software in addition to all necessary ancillary reagents. The processed plate can be read on various low-cost imagers. Applications include target gene validation; expression time courses; treatment profiles; in vitro and in vivo pharmacology; dose response curves; high throughput screening; and optimization, metabolism, safety and diagnostics. Multiplexing enables numerous different measurements to be made at one time, and generates accurate information about how a potential new drug would affect a cell or organism because many diseases, such as most cancers, are caused by the interaction of multiple factors. Additionally, multiplexing enables running controls in the same wells as the genes being tested.
