Microfluidic, Automated Electrophoresis
Experion automated electrophoresis system (Bio-Rad Laboratories) uses LabChip microfluidic technology (Caliper Life Sciences) to speed 1-D protein and RNA separation and analysis. A run typically lasts 30 minutes, including image analysis and quantitation. After basic sample preparation similar to traditional gel electrophoresis, the primed and loaded chip is inserted into the instrument, the lid is closed and analysis type selected. Results are provided in about 30 minutes. Hardware includes an automated priming station, a vortex station to mix reagents, and the automated electrophoresis station. Handling of hazardous materials is minimized. Three kits are available: protein, standard RNA, and high sensitivity RNA. Single-step molecular-weight sizing of protein samples is provided from 10 to 260 kD, while relative protein concentration is calculated for each band. Separation profiles of mRNA and total RNA samples are provided down to picogram amounts and concentrations are quantitated in nanogram amounts. Additional information on purity of total RNA is indicated by the ribosomal RNA ration number that is automatically calculated.
