Antibody Microarray

BD Clontech Ab Microarray 500 allows assaying over 500 specific proteins. A variety of proteins can be detected (both cytosolic and membrane-bound) representing a broad range of biological functions, including signal transduction, cell-cycle regulation, gene transcription, and apoptosis. Differences can be detected in protein abundance between two individual samples-cells, whole tissue, or biological fluids. The fluorescence-based procedure, which takes less than a day to complete, allows detection as little as 20 pg/ml of each protein target. A typical analysis starts with protein extraction. Proteins are extracted from cells or whole tissue in one step under non-denaturing conditions, and then labeled with green fluorescent (Cy3) and red fluorescent (Cy5) dyes. Following a simple desalting step to remove unbound dye, the labeled proteins are then incubated with the antibody microarrays. Dual-color detection method is designed so that inherent variations in dye labeling do not affect the outcome of the experiment. Thus, side-by-side comparisons accurately reflect the relative abundances of proteins in the sample-follow-up studies with Western blotting and in situ hybridization support these claims. The arrays are printed on standard-size (75 × 25 × 1 mm) glass slides, so they can be scanned with most commercially available microarray scanners.
