BacMagic DNA provides positive selection of baculovirus recombinants for protein expression in insect cells, eliminating plaque purification steps. It is an AcNPV genome with a deletion in the essential open reading frame (ORF) 1629 and an added bacterial artificial chromosome (BAC). This combination prevents non-recombinant virus from replicating in insect cells and allows the viral DNA to be propagated in bacterial cells. A compatible transfer plasmid containing the target protein coding sequence is co-transfected with BacMagic DNA into insect cells. Homologous recombination within the cells restores the function of the virus ORF1629 and the target coding sequence replaces the BAC sequence. Only recombinant baculovirus can replicate, producing a homogeneous population of recombinants. The DNA Kit includes Insect GeneJuice Transfection Reagent and a positive control plasmid.
