The Dpo4 enzyme is a thermophilic Y-family DNA polymerase that performs trans-lesion synthesis. It also allows for coupling to PCR for amplification of damaged DNA. The enzyme is purified from E. coli containing a recombinant plasmid harboring the S. solfataricus Dpo4 gene.
